PLCB4 gene
The PLCB4 gene provides instructions for making a protein involved in cell signaling that is essential for the development of the head, face, and eyes.
Normal Function
The PLCB4 gene provides instructions for making the beta 4 isoform of the protein phospholipase C. This protein is part of the phosphoinositide cycle, a signaling pathway that transmits information from the outside of the cell to the inside, ultimately influencing various cell activities.
The beta 4 isoform is critical for the development of the first and second pharyngeal arches, which form the bones and tissues of the head and face, including the jawbones, facial muscles, and ears. It is also important for the proper functioning of the retina, the light-sensitive tissue at the back of the eye.
Health Conditions Related to Genetic Changes
Auriculocondylar syndrome
Variants in the PLCB4 gene cause auriculocondylar syndrome, a disorder affecting the development of facial features, particularly the ears and the lower jaw (mandible). These genetic variants typically lead to the substitution of one amino acid for another in the beta 4 isoform of phospholipase C. These changes impair the protein's function, disrupting the cell signaling necessary for the proper development of structures derived from the first and second pharyngeal arches.
